Mitazalimab (also known as ADC-1013 or JNJ-64457107) is an investigational, humanized monoclonal antibody that functions as an agonist of the co-stimulatory receptor CD40. By binding to CD40 on antigen-presenting cells (APCs) such as dendritic cells, it mimics the action of the natural ligand CD40L, leading to activation and maturation of APCs. This enhances antigen presentation, promotes T-cell priming, and stimulates anti-tumor immune responses. Mitazalimab is being developed as an immune stimulatory therapy for the treatment of advanced solid tumors, often evaluated in combination with other immunotherapies (e.g., checkpoint inhibitors) or chemotherapy.
