Pateclizumab (also known as MLTA‑3698A or RG‑7418) is an investigational, humanized monoclonal antibody that targets lymphotoxin‑alpha (LTα, also known as TNF‑β), a cytokine belonging to the tumor necrosis factor (TNF) superfamily. LTα exists both as a soluble homotrimer and as a membrane‑bound heterotrimer with lymphotoxin‑beta (LTβ), which signals through the LTβ receptor. By binding to LTα, pateclizumab neutralizes its activity, thereby inhibiting signaling through both TNF receptors and the LTβ receptor. This blockade aims to disrupt the formation and maintenance of lymphoid‑like structures in inflamed tissues, which are thought to drive chronic inflammation in autoimmune diseases. It has been clinically evaluated for the treatment of rheumatoid arthritis (RA).
