Depatuxizumab mafodotin (also known as ABT-414) was an investigational antibody-drug conjugate (ADC) developed for the treatment of glioblastoma multiforme (GBM), the most common and aggressive primary brain tumor. It combined a humanized monoclonal antibody with a potent cytotoxic payload, designed to selectively target and kill tumor cells overexpressing a specific receptor. Clinical trials evaluated its efficacy, both as monotherapy and in combination with standard chemoradiotherapy (temozolomide), in patients with newly diagnosed or recurrent EGFR-amplified glioblastoma. Despite showing signs of activity, its development was ultimately discontinued after Phase III trials failed to demonstrate a statistically significant improvement in overall survival compared to standard of care.
