Ganitumab (development code AMG-479) was an investigational, fully human monoclonal IgG1 antibody developed for the treatment of solid tumors, including pancreatic cancer, Ewing sarcoma, and non-small cell lung cancer (NSCLC). It functioned as an antagonist of the insulin-like growth factor 1 receptor (IGF-1R). By binding with high affinity to IGF-1R on the surface of cancer and stromal cells, ganitumab blocked the binding of its natural ligands, IGF-1 and IGF-2. This blockade inhibited the receptor's tyrosine kinase activity and downstream signaling through the PI3K/AKT/mTOR and RAS/MAPK pathways, which are critical for tumor cell proliferation, survival, metastasis, and resistance to therapy.
