Letaplimab (also known as IBI-188) is a humanized IgG4 monoclonal antibody that specifically targets CD47, a cell-surface protein widely recognized as a “don’t eat me” signal. By binding to CD47, Letaplimab blocks its interaction with signal regulatory protein alpha (SIRPα) on macrophages and other myeloid cells. This blockade disrupts the inhibitory signaling that protects cancer cells from phagocytosis, thereby promoting macrophage-mediated elimination of tumor cells. It is currently under clinical investigation for the treatment of hematologic malignancies and advanced solid tumors.
