Ligufalimab (also known as AK117) is a humanized IgG4 monoclonal antibody that specifically targets CD47, a cell-surface protein known as a “don’t eat me” signal. Its IgG4 isotype is selected to minimize effector functions like antibody-dependent cellular cytotoxicity (ADCC). A key distinguishing feature of ligufalimab is its engineered binding property that does not induce red blood cell (RBC) agglutination, a common side effect associated with anti-CD47 antibodies. By blocking the interaction between CD47 and its macrophage receptor SIRPα, it selectively promotes the phagocytosis of tumor cells by macrophages. It is under clinical investigation for the treatment of advanced solid tumors and hematologic malignancies.
