Lipustobart is a humanized IgG4-kappa monoclonal antibody that specifically targets programmed cell death protein 1 (PD-1, also known as PDCD1 or CD279), an inhibitory immune checkpoint receptor. As an IgG4 isotype, it is engineered to minimize effector functions such as antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC), focusing instead on pure checkpoint blockade. By binding to PD-1 on T cells, it blocks its interaction with its ligands PD-L1 and PD-L2, thereby reversing T-cell exhaustion and restoring anti-tumor immune activity. It is being developed as an immune checkpoint inhibitor for the treatment of advanced solid tumors.
