Livmoniplimab is a humanized monoclonal antibody that targets a complex involving Leucine-rich repeat-containing protein 32 (LRRC32, also known as GARP) and latent transforming growth factor-beta 1 (TGFβ1). It specifically binds to the LRRC32-latent TGFβ1 complex on the surface of regulatory T cells (Tregs) and platelets, thereby blocking the LRRC32-mediated activation and release of mature TGFβ1. By inhibiting this localized, cell-surface activation of TGFβ1, livmoniplimab aims to selectively dampen the immunosuppressive tumor microenvironment without causing systemic TGFβ blockade. It is being investigated as an immuno-oncology agent for the treatment of advanced solid tumors, often in combination with other immunotherapies.
