Ustekinumab is a fully human monoclonal antibody targeting the p40 subunit shared by interleukin-12 (IL-12) and interleukin-23 (IL-23), two cytokines central to inflammatory and autoimmune pathways. It binds to p40, blocking interaction with their receptors and inhibiting downstream Th1 and Th17 cell activation. It is approved for the treatment of moderate-to-severe plaque psoriasis, psoriatic arthritis, Crohn's disease, and ulcerative colitis.
