Son of Sevenless homolog 1 (Sos1) is a guanine nucleotide exchange factor (GEF) that guides the exchange of Ras GDP and Ras GTP by binding to the SH3 domain of growth factor receptor binding protein 2 (Grb2), leading to the activation of ERK. Ras inhibitory peptide is a synthetic peptide containing the sequence PVPRR, which corresponds to the region in human Sos1 that interacts with the SH3 domain of Grb2. It specifically blocks the interaction between GEF and Grb2, preventing the necessary interaction for receptor tyrosine kinases (including epidermal growth factor receptors) to activate Ras.
