MCA-AVLQSGFR-Lys(Dnp)-Lys-NH2 is a FRET substrate peptide for the severe acute respiratory syndrome coronavirus main protease (SARS-CoV Mpro), containing a sequence of 10 amino acids. The substrate sequence of MCA-AVLQSGFR-Lys(Dnp)-Lys-NH2 originates from the residues P4-P5' of the N-terminal autoprocessing site of SARS-CoV Mpro with the sequence AVLQSGFRK. The enzymatic cleavage between Q4 and S5 of MCA-AVLQSGFR-Lys(Dnp)-Lys-NH2 releases the highly fluorescent 7-methoxy-coumarin (Mca) group, which is influenced by the internal quencher 2,4-dinitrophenyl (Dnp), resulting in a significant increase in fluorescence intensity.
