Necitumumab is a recombinant human IgG1 monoclonal antibody that targets the epidermal growth factor receptor (EGFR). It functions as a competitive antagonist, binding to the extracellular domain of EGFR and blocking the interaction with its ligands (e.g., EGF, TGF‑α). This inhibition prevents receptor activation and downstream signaling through the MAPK and PI3K/AKT pathways, thereby suppressing tumor cell proliferation and survival.
