Cinrebafusp alfa (PRS-343) is an investigational, high-affinity bispecific antibody developed by Pieris Pharmaceuticals using its proprietary Anticalin® platform technology. It is designed for the treatment of HER2-positive solid tumors. The drug functions by simultaneously targeting two distinct molecules: the tumor-associated antigen HER2 on cancer cells and the co-stimulatory receptor CD137 (4-1BB) on immune cells. This unique design aims to conditionally activate T cells specifically within the tumor microenvironment, thereby enhancing anti-tumor immunity while potentially minimizing systemic toxicity. The use of the Anticalin scaffold also contributes to a reduced risk of immunogenicity.
