Lucatumumab (also known as HCD122) is a fully human monoclonal antibody that targets CD40, a co-stimulatory receptor belonging to the tumor necrosis factor receptor (TNFR) superfamily. It functions as a CD40 antagonist, blocking the interaction between CD40 and its ligand CD40L (CD154). CD40 is expressed on antigen-presenting cells (e.g., B cells, dendritic cells) and some solid tumors. By inhibiting this pathway, lucatumumab aimed to suppress the survival and proliferation of malignant B cells and modulate immune responses. It was clinically evaluated for the treatment of hematologic malignancies, particularly relapsed or refractory Hodgkin lymphoma and non-Hodgkin lymphoma, but its development has been discontinued.
