制备方法

以白屈菜酸为原料,采用改进的文献方法合成4-氧代-1,4-二氢-2,6-吡啶二甲酸。具体操作如下:在0℃条件下,向反应体系中缓慢滴加425 mL 30%的氨水溶液(含NH3 41.8 g,0.21 mol),滴加过程控制在1小时内完成。滴加完毕后,将反应混合物在室温下持续搅拌48小时。反应完成后,通过减压蒸馏去除过量的氨水溶液。随后,将残余物与50 mL水混合,加入活性炭,回流15分钟进行脱色。热过滤后,收集滤液,待冷却至室温后,用37%的盐酸水溶液调节pH至1。此时,溶液中析出白色固体,经过滤收集,用冷水多次洗涤,最后在真空条件下干燥16小时,得到目标化合物4-氧代-1,4-二氢-2,6-吡啶二甲酸(13),为白色固体,产量42 g,收率98%。
