以5-氟喹唑啉-4(1H)-酮为原料合成4-氯-5-氟喹唑啉的一般步骤如下:将HATU(197 mg)、二异丙基乙胺(90 μL)、乙酸(22 μL)和N-[3-氯-4-(吡啶-2-基甲氧基)苯基]-5-[2-(甲氨基)乙氧基]喹唑啉-4-胺(150 mg)的DCM(20 mL)溶液搅拌2小时。反应完成后,用水洗涤反应液,再用盐水洗涤,最后真空浓缩。使用DCM-5%甲醇作为洗脱液,通过柱色谱法纯化残余物,得到标题化合物,为白色固体(114 mg,产率69%)。NMR光谱(DMSO-d6)δ 1.95(s,3H),3.00(s,3H),3.89(t,2H),4.48(m,2H),5.29(s,2H),7.18(d,1H),7.24(d,1H),7.35(m,2H),7.59(m,2H),7.72(dd,1H),7.85(dt,1H),7.96(d,1H),8.46(s,1H),8.58(m,1H),9.70(bs,1H);质谱m/z 478 [M+H]+。
