以1-溴-2-硝基苯(10.0 g,49.5 mmol,1.0当量)和联硼酸频那醇酯(15.08 g,59.4 mmol,1.2当量)为原料,加入乙酸钾(14.57 g,148 mmol,3.0当量)和[1,1'-双(二苯基膦)二氯化钯(II)](1.09 g,1.49 mmol,0.03当量)于二恶烷(180 mL)中。在氮气保护下,将反应混合物于80℃搅拌12小时。反应完成后,将混合物减压浓缩,得到粗产物。通过硅胶柱色谱法(洗脱剂:石油醚/乙酸乙酯,100:1至80:1)纯化,得到2-硝基苯硼酸频哪醇酯,为黄色油状物(12.0 g,收率97%)。产物经1H NMR(DMSO-d6, 400 MHz)鉴定:δ 8.17(d, J = 8.0 Hz, 1H),7.67-7.65(m, 1H),7.58-7.54(m, 2H),1.44(s, 12H)。
