一般步骤:在-78℃下,向2,4-二氯吡啶(7g,47.3mmol)的四氢呋喃(THF,70mL)溶液中缓慢加入2M二异丙基氨基锂(LDA)的THF溶液(28.4mL,56.8mmol),保持温度在-78℃并搅拌30分钟。反应完成后,用过量干冰淬灭反应混合物,随后升温至室温并继续搅拌30分钟。用1.5N盐酸(HCl)中和反应混合物后,将其用乙酸乙酯(100mL)稀释,依次用饱和盐水(2×50mL)和去离子水(100mL)洗涤。分离有机层,用无水硫酸钠(Na2SO4)干燥,减压浓缩,得到2,4-二氯烟酸(4.5g,23.44mmol,50%收率),为棕色固体。产物经1H NMR(400MHz,DMSO-d6)表征:δ 8.47(d,1H),7.74(d,1H)。
