以甲醛和3-氟苯酚为原料合成4-氟-2-羟基苯甲醛的一般步骤如下:向含有3-氟苯酚(5 mL,55.3 mmol)和无水乙腈(250 mL)的混合物中加入MgCl2(14.2 g,149 mmol)、无水三甲胺(27 mL)和多聚甲醛(11 g)。将反应混合物在回流条件下搅拌5小时,随后冷却至室温。反应通过加入5%盐酸(250 mL)淬灭,并用乙酸乙酯(100 mL × 3)进行萃取。合并的有机层依次用5%盐酸、水和盐水洗涤,经无水硫酸钠干燥后,减压浓缩。粗产物通过硅胶柱色谱纯化(洗脱剂:石油醚/乙酸乙酯=50:1至20:1),得到4-氟-2-羟基苯甲醛(5 g,产率65%),为浅黄色固体。1H NMR(400 MHz,CDCl3)数据如下:δ 11.37(d,J = 1.6 Hz,1H),9.83(s,1H),7.55-7.59(m,1H),6.65-6.75(m,2H)。
