以氢化肉桂酸内酯为原料合成4-羟基-1-茚酮的一般步骤:将氯化铝(90.0 g,676 mmol)和氯化钠的混合物在150℃下搅拌直至固体完全溶解,随后缓慢滴加氢化肉桂酸内酯(20.0 g,135 mmol)。滴加完毕后,将反应混合物升温至200℃并持续搅拌1小时。反应完成后,将混合物冷却至室温,并缓慢倒入冰水(500 mL)中,随后过滤收集粗产物。通过重结晶纯化粗产物,得到4-羟基-1-茚酮(19 g,95%收率),呈灰色固体。该化合物的结构通过以下光谱数据确认:质谱(ESI正离子模式)m/z 149.5 [M + H]+;1H NMR(400 MHz,CDCl3)δ(ppm):7.41-7.38(m,1H),7.24-7.19(m,1H),6.80-6.79(m,1H),6.78-6.77(d,1H,J = 4.0 Hz),5.46(br,1H),3.06-3.03(m,2H),2.69-2.66(m,2H)。
