向哌啶-2-酮(0.97 g,9.78 mmol)的二氯甲烷(DCM,20 mL)溶液中依次加入三乙胺(Et3N,1.36 mL,9.78 mmol)、4-二甲氨基吡啶(DMAP,0.12 g,0.978 mmol)和二碳酸二叔丁酯(Boc2O,3.20 g,14.7 mmol)。将反应混合物在室温下搅拌3小时,反应完成后,通过减压蒸馏除去溶剂。残余物通过硅胶柱色谱法纯化,洗脱剂为乙酸乙酯/石油醚(v/v = 1/7),得到1-Boc-2-哌啶酮,为浅黄色油状物(1.78 g,收率91%)。质谱(ESI,阳离子模式)m/z:144.2 [(M-C4H8)+ H]+;1H NMR(600 MHz,CDCl3):δ(ppm)3.65(t,J = 6.1 Hz,2H),2.50(t,J = 9.6, 7.2 Hz,2H),1.82(m,4H),1.52(s,9H)。
