以2,4-二氯烟醛为起始原料合成4-氯-1H-吡唑并[4,3-c]吡啶的一般步骤:在室温条件下,将中间体1(1.7g,9.7mmol)溶于二甲氧基乙烷(12ml)中,随后加入一水合肼(1.2ml,38.6mmol)。将反应混合物在75℃下搅拌反应过夜。反应完成后,将混合物浓缩至干,所得粗产物通过快速色谱法进行纯化,采用20%至100%乙酸乙酯/石油醚梯度洗脱,得到目标产物4-氯-1H-吡唑并[4,3-c]吡啶为白色固体(0.82g,产率56%)。产物结构经1H NMR(400MHz,DMSO-d6)确认:δ 7.60(d,J = 6.9Hz,1H),8.14(d,J = 6.0Hz,1H),8.32(s,1H);质谱(ES + APCI)+:m/z 154 [M + H]+。
