生产方法
c)1,1-二甲基乙基(3S)-3-(氨基甲基)-1-吡咯烷羧酸酯的合成
将1,1-二甲基乙基(3R)-3-氰基-1-吡咯烷羧酸酯(73.9 mmol)溶于乙醇(100 mL)中,置于Parr瓶中。在氮气保护下,加入Raney Nickel(73.9 mmol;约2勺Raney Nickel水溶液)。用氮气充分置换反应体系中的空气后,在60 psi的氢气压力下,将反应混合物置于Parr振荡器上反应过夜。反应完成后,在氮气保护下,将反应混合物通过硅藻土过滤,并用少量乙醇洗涤滤饼。随后,立即将滤饼用水处理。将乙醇滤液在减压下浓缩,得到标题产物,为透明油状物(71.9 mmol,收率97%)。经测定,产物的对映体比例为95:5(即>90% ee)。
采用手性HPLC(Daicel Chiralpak AD-H柱,4.6 x 150 mm;流动相:庚烷:乙醇:异丙胺=85:10:0.1;流速:1.0 mL/min;UV检测波长:215 nm)分析产物,1,1-二甲基乙基(3R)-3-(氨基甲基)-1-吡咯烷羧酸酯的保留时间为4.4分钟,1,1-二甲基乙基(3S)-3-(氨基甲基)-1-吡咯烷羧酸酯的保留时间为4.8分钟。
1H NMR(400 MHz,CDCl3)δ ppm:1.47(s,9H),1.54-1.73(m,1H),1.91-2.13(m,1H),2.28-2.52(m,1H),2.78-2.93(m,1H),2.95-3.16(m,1H),3.24-3.67(m,5H)。
