Isazophos Chemical Properties, Uses, Production

Description

Isazofos is a yellow liquid. Solubility in water is 168 mg/L (20 C). It is miscible with common organic solvents. Log Kow = 2.99. It is hydrolyzed in aqueous media: DT₅₀ values (20 C) at pH 5, 7, and 9 are 85, 48, and 19 d, respectively.

Pure product is yellow liquid. b.p. 100℃/0.1333Pa, relative density 1.22 (20 ℃), n20D1.4867, vapor pressure 1.76×10-3Pa (20 ℃). Soluble in methanol, trichlorotetramethylene and other organic solvents, solubility in water is only 150mg/L. Stable in neutral and acidic medium, easy to decompose under alkaline conditions.

Production Methods

The production of isazofos begins with the construction of a heterocyclic base, typically a substituted benzoxazole or related aromatic system, which provides the structural backbone for biological activity. This intermediate undergoes phosphorylation using reagents like phosphorochloridates or dialkyl phosphites to introduce the active phosphorothioate group, essential for its function as an acetylcholinesterase inhibitor. The reaction is carried out under controlled conditions to ensure selectivity and minimize the formation of toxic by-products. Subsequent steps may include alkylation or esterification to enhance solubility and stability. The crude product is purified through solvent extraction and distillation, yielding the active substance.

Metabolic pathway

Isazofos is metabolised in soil to its hydrolysis product 5-chloro-1- isopropyl-3-triazolol. The oxon metabolite has apparently not been identified.

Metabolism

In mammals, administered isazofos is rapidly metabolized and excreted, primarily in the urine, without accumulation in the body. The degradation route of isazofos in soil is primarily the hydrolysis mediated by microorganisms to give 5-chloro-1-isopropyl-3-triazole. The half-life in soils is about 10 d.

Toxicity evaluation

Acute oral LD₅₀ for rats is 40–60 mg/kg. Inhalation LC50 (4 h) for rats is about 0.24 mg/L air. NOEL (90 d) for rats is 2 mg/kg diet (0.1 mg/kg/d).

Degradation

Isazofos is hydrolysed more rapidly in alkaline rather than acid solution. The DT₅₀ values at pH 5, 7 and 9 were 85, 48 and 19 days, respectively (PM).
