| Synthesis | General procedure for the synthesis of 2-bromo-4-trifluoromethylpyrimidines from 4-(trifluoromethyl)pyrimidin-2(1H)-ones:[0604] Synthesis of 2-bromo-4-(trifluoromethyl)pyrimidines:[0605] To a stirred toluene solution (40 mL) of 6-(trifluoromethyl)-1,2-dihydropyrimidin-2-ol (2 g, 12.19 mmol), protected by argon gas To this solution, phosphorus bromide (5.24 g, 18.20 mmol) and N,N-dimethylaniline (156 mg, 1.20 mmol) were sequentially added at room temperature. The reaction mixture was heated to 100 °C and stirred for 3 hours. The reaction process was monitored by thin layer chromatography (TLC). Upon completion of the reaction, the reaction mixture was diluted with 50 mL of water and extracted with hexane (2 x 50 mL). The organic phases were combined, washed with 50 mL of water, dried over anhydrous sodium sulfate, and concentrated under reduced pressure to afford 2-bromo-4-(trifluoromethyl)pyrimidine as a light yellow oil (800 mg, 29% yield).
