PALMITOYL-DL-CARNITINE CHLORIDE Usage And Synthesis
UsesDL-Palmitoylcarnitine (chloride) is a long-chain acylcarnitine with both intracellular and extracellular roles. Within the cell, palmitoylcarnitine is transported into mitochondria to deliver palmitate for fatty acid oxidation and energy production. Exogenously added DL-palmitoylcarnitine alters calcium mobilization in many cell types. It also affects endothelial and epithelial tight junctions, reducing resistance and increasing permeability while decreasing cell viability.
UsesPalmitoyl-DL-carnitine chloride is a specific protein kinase C (PKC) inhibitor.
UsesPalmitoyl-DL-carnitine chloride has been used to study its effect on inflammatory cytokines and calcium (Ca2+) influx, using in vitro models of prostate cancer.
Biochem/physiol ActionsPalmitoyl-DL-carnitine chloride is known to inhibit biofilm formation mediated by Pseudomonas aeruginosa and Escherichia coli. It is a phosphokinase C inhibitor.
in vivo

Palmitoylcarnitine (10 mg/kg, s.c., once daily for 28 days) induces insulin resistance and hyperinsulinemia, leading to glucose metabolism impairment in CD-1 mice[3]. Palmitoylcarnitine (50 mg/kg, s.c., single dose) reduces insulin sensitivity and induces glucose intolerance and metabolic dysfunction in CD-1 mice[3]. Palmitoylcarnitine (50 mg/kg, i.p., single dose) increases muscle long-chain acylcarnitine content and induces insulin resistance in CD-1 mice, characterized by hyperglycemia and reduced muscle glucose uptake[3].

Animal Model:Palmitoylcarnitine (HY-126357)-induced insulin resistance CD-1 mouse model[3]
Dosage:50 mg/kg (s.c. or i.p., single dose) or 10 mg/kg (s.c., long-term administration)
Administration:Intraperitoneal injection (i.p.), single dose; Continuously released for 24 hours via a subcutaneous osmotic minipump (s.c.)., single dose Subcutaneous osmotic minipump (s.c.), once daily for 28 days.
Result:Increased muscle long-chain acylcarnitine content, elevated blood glucose levels in both fasted and fed states, and decreased muscle glucose uptake (i.p.). Induced glucose intolerance, delayed energy metabolism shift, and reduced insulin sensitivity (s.c., single dose). induced insulin resistance and hyperinsulinemia, blocked insulin-mediated blood glucose reduction, increased blood lactate levels, and impaired glucose metabolism (s.c., long-term administration).
