小鼠抗EGFR单克隆抗体

Anti-EGFR Antibody, Mouse MonoclonalMouseHumanIHC-Fr, ICC/IF, Flow CytRecombinant Human EGFR / HER1 / ErbB1 proteinThis antibody was produced from a hybridoma resulting from the fusion of a mouse myeloma with B cells obtained from a mouse immunized with purified, recombinant Human EGFR / HER1 / ErbB1. The IgG fraction of the cell culture supernatant was purified by Protein A affinity chromatography.

Immunocytochemistry/ Immunofluorescence - Anti-EGFR antibody [EGFR1] (ab30)ab30 staining EGFR (colored green) in A431 cells. The cells were fixed with 4% formaldehyde (10min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated with ab30 at 5μg/ml overnight at 4ºC. Cells were then incubated with ab150117, Goat Anti-Mouse IgG H&L (Alexa Fluor^® 488) at 1/1000 dilution (shown in green). Nuclear DNA was labelled with DAPI (shown in blue).Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

EGFR是表皮生长因子受体（HER）家族成员之一。该家族包括HER1（erbB1，EGFR）、HER2（erbB2，NEU）、HER3（erbB3）及HER4（erbB4）。HER家族在细胞生理过程中发挥重要的调节作用。 EGFR（Epidermal Growth Factor Receptor）是上皮生长因子（EGF）细胞增殖和信号传导的受体。EGFR属于ErbB受体家族的一种，该家族包括EGFR (ErbB-1),HER2/c-neu(ErbB-2),Her 3(ErbB-3) 和Her 4(ErbB-4)。EGFR也被称作HER1、ErbB1，突变或过表达一般会引发肿瘤。EGFR是一种糖蛋白，属于酪氨酸激酶型受体，细胞膜贯通，分子量170KDa。EGFR位于细胞膜表面，靠与配体结合来激活，包括EGF和TGFα（transforming growth factor α）。激活后，EGFR由单体转化为二聚体，尽管也有证据表明，激活前也存在二聚体。小鼠抗EGFR单克隆抗体是一类可以特异性结合EGFR的单克隆抗体，主要用于检测EGFR的Western Blot、IHC-P、IF、ELISA、Co-IP等多种免疫学实验。双抗体夹心法测定标本中EGFR水平。用纯化的Anti-ACTA2抗体包被微孔板，制成固相抗体，往包被单抗的微孔中依次加入EGFR，再与HRP标记的EGFR抗体结合，形成抗体-抗原-酶标抗体复合物，经过彻底洗涤后加底物TMB显色。TMB在HRP酶的催化下转化成蓝色，并在酸的作用下转化成最终的黄色。颜色的深浅和样品中的EGFR呈正相关。用酶标仪在450nm波长下测定吸光度（OD值），通过标准曲线计算样品中EGFR浓度。
