Background Description

Probucol is a lipid-lowering drug that regulates blood lipids and exhibits antioxidant effects by reducing cholesterol synthesis and promoting cholesterol breakdown. It is commonly used to treat hypercholesterolemia and prevent atherosclerosis. Impurity 26, a process-related impurity in its synthesis, may originate from oxidative coupling side reactions of phenolic raw materials or incomplete thio-reactions. Its disulfide bond, tert-butyl group, and phenolic hydroxyl group may affect the drug's stability, antioxidant activity, and safety. Since lipid-lowering drugs are taken long-term, strict control of impurities is directly related to patient safety, making the study of this impurity an important part of ensuring drug quality.

Research Status

  • Detection Technology:UPLC-MS/MS technology, combined with a C18 column (1.7μm) and gradient elution with 0.1% formic acid - acetonitrile, achieves impurity separation within 10 minutes, with a detection limit as low as 0.003 ng/mL for high-precision trace impurity detection.

  • Formation Mechanism:Studies show that this impurity is formed by the coupling reaction of 2,4-di-tert-butylphenol under the action of an oxidant (such as hydrogen peroxide) and treatment with a thio-reagent (such as sodium sulfide). Optimizing the dosage of oxidant and thio-reagent and the reaction pH can effectively inhibit side reactions.

  • Safety Evaluation:In vitro cytotoxicity experiments show that the IC₅₀ of this impurity against HepG2 cells is 192.6 μM (Probucol IC₅₀ = 10.3 μM). Although the toxicity is lower than that of the main drug, its content in drugs still needs to be strictly controlled. Currently, long-term stability tests are being carried out to systematically study its degradation characteristics and potential risks under high temperature, high humidity, and light conditions
