| In vitro | Method: MCF10A reporter cells were treated with SID 3712249/compound 1 at 20 nM for 48 h, and precursor and mature miR-544, BMI1/mTOR 3′-UTR reporter activity and downstream transcripts were measured. MDA-MB-231 and MCF-7 cells were exposed to 20 nM for 5 days under hypoxia or normoxia. Result: SID 3712249 caused accumulation of pre-miR-544, reduced mature miR-544 and disrupted miR-544-dependent reporter repression. It induced apoptosis in MDA-MB-231 and MCF-7 cells under hypoxia and sensitized cells to 5-fluorouracil, while having little effect on normoxic survival.[1] |
| In vivo | Method: MDA-MB-231-GFP-luc cells were implanted into NOD/SCID mice. Compound 1 was evaluated either after in-vitro pretreatment of tumor cells or by intraperitoneal administration 24 h after implantation; tumor bioluminescence and resected tumors were analyzed over 21 days. Result: Compound 1 inhibited tumor growth at a lower effective dose than the miR-544 antagomir and altered miR-544 precursor/mature levels and hypoxia-associated transcripts in excised tumors.[1] |
