| Description | Synucleozid hydrochloride inhibits the translation of the intrinsically disordered protein α-synuclein by targeting its structured mRNA. Synucleozid hydrochloride targets the IRE structure and inhibits SNCA translation in cells, and can be used to study Alzheimer's disease. |
| In vitro | b>METHODS: The affinity of Synucleozid (100 nM-100 μM) for its putative binding site A in the IRE was measured by using a fluorescent adenine analog to 2-aminopurine (2-AP). RESULTS: Synucleozid reduced 2-AP emission with an EC50 of 2.7 ± 0.4 μM; importantly, restoration of 2-AP emission was observed as a function of the concentration of unlabeled SNCA IRE RNA (RNA-0), providing a competitive Kd value of 1.5 ± 0.3 μM.
