The ZBTB16 antibody is a crucial tool for studying the zinc finger and BTB domain-containing protein 16 (ZBTB16), also known as promyelocytic leukemia zinc finger (PLZF). ZBTB16 is a transcription factor involved in cell differentiation, proliferation, and apoptosis. It contains an N-terminal BTB domain for protein interactions and C-terminal zinc finger motifs for DNA binding. Initially identified through its association with acute promyelocytic leukemia (APL), ZBTB16 forms a fusion gene with RARA in rare APL cases due to chromosomal translocations (t(11;17)), impacting myeloid differentiation. Beyond oncology, ZBTB16 regulates stem cell maintenance, immune cell development (e.g., natural killer T cells), and organogenesis (e.g., limb, brain).

ZBTB16 antibodies are widely used in techniques like Western blotting, immunohistochemistry (IHC), and chromatin immunoprecipitation (ChIP) to detect protein expression, localization, and DNA-binding activity. These antibodies aid in exploring ZBTB16's role in transcriptional repression, often mediated by recruiting chromatin-modifying complexes like Polycomb repressors. Commercial ZBTB16 antibodies are typically raised in rabbits or mice, with clones validated for specificity using knockout controls.
