The GRIA2 antibody targets the GluA2 subunit of the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor, a critical ionotropic glutamate receptor in the central nervous system. Encoded by the GRIA2 gene, GluA2 plays a pivotal role in mediating fast synaptic transmission, synaptic plasticity, and learning/memory processes. A hallmark feature of GluA2 is its regulation of calcium permeability: most AMPA receptors containing GluA2 are impermeable to calcium due to RNA editing at the Q/R site, preventing excitotoxicity. Dysregulation of GRIA2 expression or function has been implicated in neurological disorders such as Alzheimer's disease, epilepsy, ischemic stroke, and neuropathic pain.

GRIA2 antibodies are widely used in neuroscience research to study receptor localization, trafficking, and post-translational modifications. They enable detection of GluA2 in techniques like Western blot, immunohistochemistry, and immunofluorescence, aiding investigations into synaptic changes under pathological conditions. Additionally, autoantibodies against GRIA2 have been identified in autoimmune encephalitis and paraneoplastic syndromes, where they disrupt synaptic signaling, leading to cognitive deficits or seizures. These clinical associations highlight the antibody's dual role as both a research tool and a biomarker. Commercially available GRIA2 antibodies are typically raised against specific epitopes, with validation emphasizing specificity for GluA2 over other AMPA receptor subunits (GluA1. GluA3. GluA4).
