SERPINB4. a member of the serine protease inhibitor (serpin) superfamily, is encoded by the SERPINB4 gene located on chromosome 18q21.3. It is also known as squamous cell carcinoma antigen 2 (SCCA2) and shares structural homology with SERPINB3 (SCCA1), both clustered in the same genomic region. Primarily expressed in squamous epithelial cells, SERPINB4 inhibits proteases like kallikreins and cathepsins, playing roles in immune regulation, inflammation, and tissue remodeling. Dysregulation of SERPINB4 is linked to squamous cell carcinomas (e.g., cervical, lung, head/neck cancers), where its overexpression correlates with tumor progression, metastasis, and apoptosis resistance. It is also implicated in inflammatory skin disorders such as psoriasis and atopic dermatitis, contributing to abnormal keratinocyte differentiation and immune activation.

SERPINB4 antibodies are critical tools for detecting this protein in research and diagnostics. They are used in techniques like Western blotting, immunohistochemistry (IHC), and ELISA to study SERPINB4 expression patterns in tissues or biofluids. These antibodies aid in exploring its role as a biomarker for cancer prognosis or therapeutic targeting. However, challenges include potential cross-reactivity with SERPINB3 due to high sequence similarity, necessitating rigorous validation.
