髓系细胞表面抗原CD33(亦称为SIGLEC3、Siglecs(唾液酸结合免疫球蛋白样凝集素)及GP67)是一种单次跨膜的I型膜蛋白,隶属于免疫球蛋白超家族及SIGLEC(唾液酸结合免疫球蛋白样凝集素)家族。人源CD33/Siglec-3 cDNA编码364个氨基酸的多肽链,包含疏水信号肽、N端Ig样V型结构域、一个Ig样C2型结构域、跨膜区及胞质尾部。CD33/Siglec-3通常被认为具有髓系特异性,但也可在某些淋巴样细胞上表达。在免疫应答中,当配体诱导酪氨酸磷酸化后,CD33/Siglec-3可通过其SH2结构域招募胞质磷酸酶,作为抑制性受体通过去磷酸化信号分子来阻断信号传导。CD33/Siglec-3能够诱导急性髓系白血病细胞发生凋亡。
