Pertuzumab is a humanized monoclonal antibody that targets human epidermal growth factor receptor 2 (HER2), specifically binding to subdomain II of the HER2 extracellular domain. This binding inhibits HER2 dimerization with other HER family members (especially HER3), thereby blocking ligand‑dependent signaling pathways (e.g., PI3K/AKT, MAPK) that drive tumor cell growth and survival. Pertuzumab is approved in combination with trastuzumab and chemotherapy for the treatment of HER2‑positive breast cancer—both in the neoadjuvant/adjuvant setting for early‑stage disease and in the first‑line metastatic setting—as well as in combination with trastuzumab and docetaxel for HER2‑positive metastatic gastric or gastroesophageal junction adenocarcinoma. It is administered via intravenous infusion.
