Itolizumab is a humanized, recombinant monoclonal antibody that uniquely targets the CD6 receptor on T cells. It functions as an immune modulator by binding to the SRCR-1 domain of CD6, thereby interfering with the CD6-ALCAM (Activated Leukocyte Cell Adhesion Molecule) interaction. This action inhibits T-cell activation, proliferation, and migration to inflammatory sites. Itolizumab is notable for being one of the first anti-CD6 antibodies approved for therapeutic use, specifically in India for moderate to severe chronic plaque psoriasis. It is also being investigated for other autoimmune conditions and cytokine storm-associated complications, such as in severe COVID-19.
