Methyl levulinate synthesis

14synthesis methods
The acid was converted to the ester by boiling in methanol (0.5 M solution) in the presence of concentrated sulfuric acid (500 μL). The reaction was followed by TLC (heptane/EtOAc 2:1). When only trace amounts of acid remained, the reaction mixture was co

Reaction Conditions:

with sulfuric acid at 50; for 5 h;

Steps:

Methyl 4-oxopentanoate (2a)

General procedure: Sulfuric acid (0.1 ml) was added to a solution of levulinic acid (0.782 g, 7.0 mmol) in methanol (10 ml) and the mixture was heated at 50° C for 5 hours. The solution was evaporated under reduced pressure, and the residue was partitioned between saturated aqueous solution of NaHCO3 and ethyl acetate. The organic layer was separated, extracted with brine solution and dried over anhydrous MgSO4. Evaporation under reduced pressure afforded compound 2a (0.901 g, 99%). 1H NMR (400 MHz, CDCl3) δ 3.68 (3H, s, -OCH3), 2.76 (2H, t, J = 6.4 Hz, H-2), 2.58 (2H, t, J = 6.4 Hz, H-3), 2.20 (3H, s, -CH3).
