一般步骤:在搅拌条件下,将POCl3(60 mmol,6 mL)缓慢滴加至预先冷却至0℃的干燥DMF(30 mL)中,制备Vilsmeier-Haack试剂。随后,将反应混合物在0℃下继续搅拌10-15分钟。将4-甲基吲哚(10 mmol)溶解于DMF(5 mL)中,所得溶液加入上述制备的Vilsmeier-Haack试剂内。将反应混合物升温至35℃,持续搅拌1小时。反应完成后,冷却至室温,依次加入冰水(6 mL)和30% NaOH水溶液(13 mL),随后将混合物加热回流20分钟,再冷却至室温。用二氯甲烷(20 mL×3)萃取反应混合物,合并有机相,用无水Na2SO4干燥。减压浓缩除去溶剂,所得粗产物通过快速柱色谱法纯化,采用15-25%丙酮/石油醚(沸程60-90℃)作为洗脱剂,得到目标产物4-甲基吲哚-3-甲醛。
