通用程序:将三氯氧磷(POCl3,1.73 mL,18.6 mmol)缓慢滴加到搅拌并冰浴冷却的N,N-二甲基甲酰胺(DMF,5.0 mL)中。反应混合物在1-5℃下搅拌20分钟后,缓慢加入2-甲基吲哚(15.5 mmol)的DMF(5.0 mL)溶液。将所得反应混合物逐渐升温至35℃,并在该温度下维持40分钟。随后将混合物冷却至室温。向混合物中加入冰(约10 g),随后加入5M氢氧化钠(NaOH)溶液(31 mL,155 mmol)。反应混合物在95℃下加热30分钟后,冷却至室温。再次向混合物中加入冰(约10 g),并将所得反应混合物搅拌30分钟。通过过滤收集2-甲基吲哚-3-甲醛,并用蒸馏水洗涤数次。
