以2-氯吡啶为原料合成2-肼吡啶的一般步骤:向2-氯吡啶(20g,0.176mol,1当量)的水合肼(200mL,10体积)溶液中,在100℃下搅拌反应48小时。通过薄层色谱(TLC,展开剂比例为8:2的乙酸乙酯和甲醇)监测反应进程,确认原料完全消耗后,将反应混合物用水(200mL)稀释,随后用乙酸乙酯(5×500mL)进行萃取。合并有机相,用无水硫酸钠(Na2SO4)干燥,减压浓缩除去溶剂,得到目标产物2-肼吡啶(15.0g,收率78%),为红色油状物。产物经1H NMR(300MHz,CDCl3)表征:δ8.14(1H,d,J = 3Hz,Ar-H),7.51-7.45(1H,m,Ar-H),6.71-6.66(2H,m,Ar-H),5.78(1H,brs,-NH),3.81(2H,brs,-NH2)。LCMS分析:计算值C5H7N3[M+H]+为109.13,实测值为110.1。
