以2-(羟甲基)丙烯酸甲酯为原料合成(S)-(+)-3-羟基-2-甲基丙酸甲酯和(-)-甲基-D-BATA-羟基异丁酸酯的一般步骤:K.3。不对称催化氢化的典型方法如下:将[Rh(COD)L*]BF4(0.005 mmol,1 mol%)和底物(0.5 mmol)溶解于干燥的溶剂(7.5 mL)中,然后将该溶液转移至不锈钢高压釜内。密封高压釜后,先用氢气进行吹扫,再用氢气加压。在室温下搅拌反应混合物16小时后,缓慢释放氢气至大气压,并将反应溶液转移至圆底烧瓶中。利用旋转蒸发器去除溶剂,得到粗产物。该粗产物通过硅胶柱色谱进行纯化,得到氢化产物。产物的对映体过量值通过手性柱HPLC测定。使用配体(1-48)或(1-49)的铑配合物进行不对称催化氢化的结果分别列于表9和表10中。表9展示了配体(1-48)的铑配合物在不同条件下的不对称催化氢化结果,包括底物、溶剂、压力、转化率和对映体过量值。表10则展示了配体(I'-49c)的铑配合物在相同条件下的不对称催化氢化结果。
