Production Methods
The industrial synthesis of pretilachlor begins with the formation of 3-propoxypropionaldehyde, which is prepared by reacting sodium n-propoxide with 2-chloroacetaldehyde dimethyl acetal. This intermediate is then subjected to reductive amination with 2,6-diethylaniline in the presence of a hydrogenation catalyst under controlled pressure and temperature to yield the corresponding aminoether. The final step involves acylation of the aminoether with chloroacetyl chloride in a biphasic system using sodium hydroxide and toluene, forming the active pretilachlor compound. These reactions are carefully managed to ensure high yield and purity, with purification typically achieved through crystallisation or solvent extraction.
