The camptothecin nitride (500 mg, 1.37 mmol) prepared in Example 1 (A) was dissolved in a mixed dioxane-acetonitrile-water solvent (5:4:1, 500 mL, by volume). Concentrated sulfuric acid (1 mL) was added to this solution and the reaction was subsequently carried out under light conditions for 90 min. After completion of the reaction, the mixture was concentrated under reduced pressure and diluted with deionized water (1 L). The precipitate was collected by filtration and dried to give the crude product. The crude product was washed with methanol and purified by silica gel column chromatography, resulting in 441 mg of 10-hydroxycamptothecin in 88.2% yield. The obtained product was compared with the standard isolated from the hi-tree (Camptotheca acuminata Nyssaceae) by various spectral analysis methods to confirm its structure. | | References | [1] Patent: US4473692, 1984, A |
