Synthesis Example 19: Synthesis of Intermediate 16 In a dry reaction flask, 3.08 g (13 mmol) of 2,6-dibromopyridine, 1.23 g (10 mmol) of pyridine-4-boronic acid, 0.58 g (0.5 mmol) of tetrakis(triphenylphosphine)palladium (Pd(PPh3)4) and 5.53 g (40 mmol) of potassium carbonate (K2CO3) were added. The above mixture was dissolved in 50 mL of THF/H2O (2:1 by volume) solvent mixture. The reaction system was stirred at 80°C for 5 hours. After completion of the reaction, the mixture was cooled to room temperature and extracted three times with 60 mL of diethyl ether. The organic layers were combined, dried with anhydrous magnesium sulfate, filtered and concentrated under reduced pressure to remove the solvent. The resulting residue was recrystallized by a solvent mixture of dichloromethane and n-hexane to give 1.74 g of white solid product (yield: 74%), Intermediate 16.The structure of the compound was confirmed by high resolution mass spectrometry (HR-MS), with a calculated value of 233.9793 and measured value of 233.9796 for C10H7BrN2.
