Preparation

Under an argon atmosphere, a trace amount of iodine was added to a mixture of magnesium (434 mg) and THF (2 mL). The mixture was heated to 60 °C, and a solution of 4-bromo-2-chloro-1-fluorobenzene (3.58 g) in THF (6.5 mL) was added dropwise (the temperature was maintained at 65 °C during the addition). The mixture was stirred at 60 °C for 30 minutes and then cooled to room temperature to give 3-CHLORO-4-FLUOROPHENYLMAGNESIUM BROMIDE.

ApplicationMagnesium 3-chloro-4-fluorophenyl bromide can be used as a pharmaceutical synthesis intermediate, such as in the preparation of pyrrolidine derivatives like (2R,5S)-1-(tert-butoxycarbonyl)-5-(3-chloro-4-fluorophenyl)pyrrolidine-2-carboxylic acid ethyl ester. This compound has antagonistic activity against the calcitonin gene-related peptide (CGRP) receptor and can be used to treat various diseases related to the CGRP receptor.
