Production MethodsThe commercial production of fluorodifen follows a concise two-step nucleophilic aromatic substitution. First, 2-chloro-5-(trifluoromethyl)nitrobenzene is prepared by nitration of 4-chlorobenzotrifluoride with mixed acid, yielding the 2-nitro isomer after steam distillation. This intermediate is then reacted with 4-nitrophenol sodium salt displacing chloride to form the diaryl ether linkage; the mixture is diluted with water, acidified, and the crude product filtered, washed, and recrystallised from ethanol to technical-grade fluorodifen.
Mechanism of actionInhibits non-cyclic electron transport
Safety ProfileMildly toxic by ingestion. An eyeirritant. Used as an herbicide. When heated todecomposition it emits very toxic fumes of NOx and Fí.
Metabolic pathwayPhotopysis products of eight substituted diphenyl ethers (preforan, nitrofen, MC-338, MC-4379, MC-3761, MC-5127, MC-6063, and MC-7181) irradiated by a Raryonette reactor in solutions of water, cyclohexane, and methanol indicates that the major reaction pathways of these biphenyl ethers include reductive dehalogenation, decarboxyalkylation, reduction of nitro substituents to amines, and cleavage of the ether linkage to yield phenols.
