以4-溴-2-氟苯胺和7-苄氧基-4-氯-6-甲氧基喹唑啉为原料合成7-(苄氧基)-N-(4-溴-2-氟苯基)-6-甲氧基喹唑啉-4-胺的一般步骤:将6.5M的氯化氢(2.54mL)加入到含有4-溴-2-氟苯胺(4.51g,15.0mmol)和7-苄氧基-4-氯-6-甲氧基喹唑啉(2.40g,16.5mmol)的2-丙醇(160mL)溶液中。随后,将反应混合物加热至回流状态并保持2小时。反应完成后,将混合物冷却至室温,过滤收集析出的固体产物。用2-丙醇和乙醚依次洗涤固体,最后在真空条件下干燥过夜,得到目标化合物7-(苄氧基)-N-(4-溴-2-氟苯基)-6-甲氧基喹唑啉-4-胺(7.9g,产率94%),为白色固体。
