Propargyl-PEG4-Tos Usage And Synthesis
DescriptionPropargyl-PEG5-Tos is a PEG linker containing a tosyl group and a propargyl group. The hydrophilic PEG spacer increases solubility in aqueous media. The tosyl group is a very good leaving group for nucleophilic substitution reactions. The propargyl group can react with azide-bearing compounds or biomolecules via copper catalyzed azide-alkyne Click Chemistry to yield a stable triazole linkage.
Biological ActivityPropargyl-PEG4-Tos is a PROTAC linker belonging to the PEG class and can be used to synthesize a series of PROTAC molecules. It is a cleavable ADC linker for the synthesis of antibody-drug conjugates (ADCs).

PROTACs contain two different ligands connected by a linker; one is a ligand for an E3 ubiquitin ligase and the other is for the target protein. PROTACs exploit the intracellular ubiquitin-proteasome system to selectively degrade target proteins.
ADCs are comprised of an antibody to which is attached an ADC cytotoxin through an ADC linker.
