| Synthesis | General procedure for the synthesis of 2-morpholino-4-hydroxypyrimidine from 2-methylthio-4-pyrimidinone (4.0 g, 0.0281 mol) and morpholine (3.05 g, 0.035 mol): 2-methylthio-4-pyrimidinone was mixed with morpholine, heated to 145°C and maintained for 2 hours. After completion of the reaction, it was cooled to room temperature. White needle-like crystals (2.0 g) were obtained by ethanol crystallization. Subsequently, a second batch of crystals of about 0.50 g was formed in a total yield of 49%. The product was detected by mass spectrometry showing M + H = 181.1. 1H NMR (CDCl3) data were as follows: δ 12.1 (broad peak, 1H), 7.85 (double peak, 1H), 5.79 (double peak, 1H), and 3.75 (multiple peaks, 8H). | | References | [1] Patent: WO2005/28444, 2005, A1.
