Step 5. Synthesis of 7-bromo-2-chloroquinazoline: A suspension of 7-bromoquinazolin-2(1H)-one (0.5 M) in phosphoryl chloride was placed in an oil bath, heated to 110 °C and maintained for 1 hour. After completion of the reaction, the mixture was cooled to room temperature. Subsequently, the volatiles were removed under reduced pressure. The residue was ground with ice water and the resulting solid was collected by filtration and dried in air to afford the target product 7-bromo-2-chloroquinazoline in 65% yield. The product was analyzed by electrospray mass spectrometry (ES/MS) and showed m/z of 243/245 (MH+).
